Health Ministry Proposes Stricter Microbial Limits for Grade A Clean Rooms Under Schedule

A proposed revision has been made by the Union Ministry of Health and Family Welfare in order to provide more stringent microbial contamination limits for the Grade A clean zones where sterile pharmaceutical products are manufactured. This new amendment to the Schedule M in the Drugs Rules, 1945 seeks to match India’s manufacturing requirements with that of WHO’s and provide protection to sterile drugs.
This proposed amendment has been issued through G.S.R. No. 953(E) of October 28, 2026, which seeks to make changes to the microbial contamination limits recommended in Part II of the Schedule M. The ministry notified about this amendment on November 4.
Grade A Clean Rooms to Follow a ‘No Growth’ Standard
The Grade A area is one of the most significant zones in pharmaceutical manufacturing operations. This is an environment where manufacturers work on exposed sterile products, containers, and sterilised parts.
As per the new amendment, the Grade A area will have a strict ‘no growth’ criteria in four monitoring zones including air, settle plates, contact plates, and glove prints.
This amendment aims at eliminating any gap between current Schedule M rules and the recommended WHO guidelines under Technical Report Series (TRS) No. 1044, Annexure 2 on good manufacturing practices for sterile pharmaceutical products.
Microbial contamination is expressed in terms of colony forming units (CFUs), an accepted measurement of microbial content of a sample. The new Grade A area criterion implies that no microbial growth should be detected in the above-mentioned monitoring samples.
Any Microbial Growth Will Trigger an Investigation
One of the features of the amendment is that microbial growth found in any area categorized as Grade A should be considered grounds for further investigation.
With this amendment, more attention would be drawn to discovering the source of contamination than to the actual microbial levels that are deemed to be too low. It would be up to manufacturers to determine the source of such growth – it might be related to cleaning procedures, personal hygiene of employees, machinery, air handling systems, etc.
Such an amendment highlights the role of GMP.
Industry Stakeholders Invited to Submit Suggestions
The ministry has solicited any objections or suggestions from the concerned stakeholders and citizens of the nation. The new rules will be looked into after thirty days from the day of availability of relevant Gazettes notifications to the public. The government will go through all the suggestions received within this time frame and then make a decision regarding the final rules.
Suggestions may be made to the Under Secretary (Drugs), Ministry of Health and Family Welfare, Government of India, Kartavya Bhawan-1, New Delhi. One may also send his/her response through the following e-mail address: [email protected].
This move is one of the steps taken by India to tighten the quality control system for medicines and harmonize its manufacturing system with the international standards. Once these limits get finalized, manufacturers will have to take full responsibility of maintaining a Grade A contamination-free environment.
